What about necrotrophic pathogens? Well, ppGpp is required for the resistance against these pathogens (i.e: Botrytis cinerea)!!!!

Also, we found that ppGpp controls photosynthetic efficiency during pathogen infection. Moreover, we demonstrated that pathogen T3SS is required for ppGpp-mediated downregulation of photosynthetic efficiency.

so, we decided to carry on and infect (with Pseudomonas) plants with high and null levels of ppGpp. High levels of ppGpp make the plants more sensitive to this pathogen while null plants are more resistant. We confirmed this by using a line with an inducible ppGpp synthase👇

Surprisingly, SA pathway, but not SA itself, is required for the induction of ppGpp biosynthesis genes during pathogen infection.😨

Ok, so ppGpp deregulates immunity-linked genes, but is this molecule produced under infection conditions? By infecting plants with Pseudomonas (hemi-biotrophic) and Botrytis (necrotrophic), we found that ppGpp biosynthesis genes are induced alongside with JA and SA genes.

As every crisis means opportunity time, we did find that plant alarmone induces a deregulation of nuclear-encoded immunity-related genes, in normal conditions!

At the very beginning of my PhD we decided to check whether this molecule can modify the light- and dark-related transcriptomes. Well, guess what, no 😪. The main hypothesis of my PhD had fallen apart at that point.
